Evening All! Its time for the March 2016 1500 thread round up The end of March already and we’re back to lovely evening runs in the sunshine. Or windswept by storms depending on the whims of the weather! The target to reach by the end of March is 372.95 miles to be on track for 1500 miles by the end of the year. The highest mileage this month was clare1976 with 727.36 miles Well done! Closest to target mileage is intrepid explorer ChrisHB with 373.05 Excellent work! This month’s round up theme is special stains used in histology to assist in diagnosis. I started my career in a histology lab, usually finishing work with a multi-coloured lab coat! The origin of Histology dates back to the 1700s with the increased use of the microscope to investigate illness led scientists to realise that the body’s structures are composed of different substances. Being able to identify those substances can aid diagnosis, causes of disease and also how they can be treated. Histochemistry took off in the 1950s, with many of the techniques developed still in use today. I’ve chosen special stains that relate to running in different ways, hope you find it interesting! Special stain: Caljal’s gold impregnate for astrocytes. anatomyatlases.org The most important muscle that a runner uses – their brain! Getting the right mental approach can make a massive difference. Astrocytes can fuel neurons with glucose during periods of high rate of glucose consumption and glucose shortage. Recent research suggests there may be a connection between this activity and exercise: clare1976 727.36 zuistu 662.62 FenlandRunner 613.12 Mrs Jigs (Luverlylegs) 606.86 BigEvilC 595.72 Jono. 569.99 jabberknit 560.24 Techthick 546.38 Uyuni 541.46 mattleydon 534.91 Ron Burgundy 530.15 Special stain: Grimelius for Adrenal Medualla (Argyrophilic cells) tandfonline.com The adrenal medulla—the inner part of the gland—produces hormones, such as adrenaline (which helps your body react to stress). The start of a race or the sprint finish will certainly see the adrenal gland working hard. The silver impregnates the tissues and leaves a brown-black deposit showing the active granules in the adrenal gland: Pikelet 515.95 jennyh 513.10 maT.T. 476.61 StuHolmes 445.56 minardi 441.3 run-forest-run 438.51 MzG 431.51 Ndwdave 429.89 Huntsman 426.03 Wooters 422.12 Special stain: Massons Trichrome image.slidesharecdn.com Connective tissue – collagen fibres are found in large quantities through the body. They can form large bundles of fibres clumped together to form structures of great tensile strength such as tendons. Runners are particularly interested in one tendon – the Achilles, which is prone to injuries. Masson’s trichrome is a bright, colourful special stain where the collagen stains green, and muscle red: RichHL 412.02 RunningInCircles 410.08 StuartM 406.66 DocMoye 405.57 Cog Noscensme AHA 400.56 FRU (KeepTheFaith) 398.81 Wazelle 383.12 ChrisHB 373.05 HappyG(rrr) 372.25 Unity 360.22 Special stain: myofibrillar ATPase image.slidesharecdn.com This is proper histochemistry, which always foxed me when I was a student! Muscle fibres (can be slow, fast – fatigue resistance, fast fatiguable). The different types of muscle fibres can be identified by using chemical reactions within the tissue. Enzymes present within frozen sections of muscle fibres can be chemically reacted with certain products to visualise the enzyme activity. Using myofibrillar ATPase fast and slow muscle fibres can be identified, as the fast muscle fibres react quicker with ATP and appear dark. People have different % of fast and slow muscle fibres but they all need to be trained: _andy 359.78 Shadow 344.7 Watford Wobble 338.61 Flatlander 334.08 dafinchi 319.05 Fizz 315.56 Neilio 313.23 cerid 309.52 Toks 306.54 Steady as she goes 290.4 Special stain: Safranin-O spandidos-publications.com Bone and cartilage We couldn’t be runners without the skeleton and it’s lubricants for our muscles to work off! Hopefully none of us will suffer a broken bone. The structure of bone and its formation is quite amazing to see. Histological examination of bone is quite challenging. It either requires mounting in plastic (methylmethacrylate which smells terrible) or demineralising first (which can take weeks but not as long as plastic). Safranin-O can be used to identify cartilage – this stains orange to red, the nuclei stains black and the background green: pielips 286.88 That man from March 270.54 mole_thing 260.23 Garfield 245.55 IanS 205.85 Pedro_Perez 190.73 Maclennane 171.2 Keefy Beefy 85.96 JovialGnome 78.75 kstuart 27.97 Well done to everyone, all on the 1500 thread. I may not post everyday but I do read everyone’s posts – you all inspire me. Such a great bunch of people and amazingly friendly. Keep it up! |

I'm pleased I'm in a group that foxed MzG as as student. I am wholly slow twitch muscles as will be ably demonstrated on Sunday. MzG is a lovely person as I found out at Ely10k this year. If you need a cheer she's the girl you need. Thanks MzG. |
